過程:第2屆的RNAi Asia年度會議(2010年11月15 – 16日, 新加坡)及展示會由the Genome Institute of Singapore (GIS)的Stem Cell and Developmental Biology部門senior group leader Dr. Bing Lim擔任主席。此外,該會議由GIS與新加坡Agency for Science, Technology and Research (A*STAR)的生物醫學研究機關共同主辦,本人應 Dr. Bing Lim 之邀請於會中演講。


Identification of microRNA Functional Effectors using Caenorhabditis elegans


In humans, let-7 regulates the RAS oncogene and defective expression of let-7 is associated with lung and breast cancers. In C. elegans, let-7 regulates heterochronic genes, including lin-41, let-60/RAS and hbl-1/lin-57, and controls the timing of terminal differentiation decisions. Dysregulation of let-7 results in irregular hypodermal and vulval development, which can be used as markers in RNAi screening of let-7 functioning effectors. We searched candidates from complexes containing let-7 and/or Argonaute protein ALG-1, purified by affinity selection and immunoprecipitation, and used feeding RNAi to reduce their gene expression in let-7(n2853) mutant. Reduced expression of the let-7 functioning effectors suppressed the aberrant vulva and hypodermis development phenotypes of let-7(n2853) mutant animals. We have identified the DEAD-box RNA helicase DDX47 and ribosomal proteins, RACK1 and RPS14, as functioning effectors of the let-7. These results lead to novel mechanisms in microRNA function, all with the potential to be involved in human cancer.
